臺灣金線連被廣泛應用於治療糖尿病、高血壓及腫瘤等病症,甚具醫療價值,由於天然生產之金線連不敷藥用所需,如以傳統的播種法繁殖,種子也很難發芽,因此利用組織培養技術提高種子萌芽率,大量繁殖此一重要的藥用植物種苗,是本研究之目的。試驗結果顯示含有1/2MS基本鹽類配合1g/1HYPONeX及1g/1 peptone之培養基,有助於金線連種子之萌芽,播種90天後,胚萌芽並突破種皮形成根莖之百分率為17%,稍高於對照組14.6%;此外,以較低成熟度之種子(62.5%-87.5%)播種,約有10.5-18.5%之萌芽率。播種前如以3%過氧化氫溶液預處理這些幼嫩種子3分鐘,會導致種胚死亡而無法形成根莖;若將較老熟種子(成熟度87.5%以上)以3%過氧化氫溶液預處理,則有高達35%之種子能萌芽並形成根莖;可能因3%過氧化氫溶液預處理可洗除金線連老熟種子種皮所含之萌芽抑制物質,進而提高其發芽率。
Anoectochilus formosanus Hayata has been widely used in Taiwan for treating many diseases such as diabetes, high blood pressure, and tumors, This medicinal herb is short in supply due to over-harvest of the naturally-grown plants and poor germination of the seeds. The purpose of this study was to explore ways to improve the seed germination rate through tissue culture techniques in order to establish a mass-propagation system of this precious plant species. Experimental results indicated that medium containing 1/2 MS basic salts with 1 g/l each of HYPONeX and peptone slightly increased the seed germination rate as compared with the control treatment (17.0% vs. 14.6%). Culture of the less-matured seeds (62.5%-87.5% in maturity) resulted in germination rates of 10.5-18.5%. Pretreatment of the less-matured seeds with 3% H2O2 for 3 min was detrimental to the viability of embryos and no rhizome was produced. However, H2O2 pretreatment of mature seed (over 87.5% in maturity) increased the germination rate to 35.0% with vigorous rhizome development. It was presumed that pretreatment of mature seed with 3% H2O2 for 3 min would remove substances inhibitory to germination from the seed-coat and thereby increase the seed germinatioin rate.
