且且圖分析放射線照射及8 S 0 處理對於多形性神經膠母瘤細胞( g l i o b l a sto m a multiforme, G 8M) 內GS H 含量之影響。 拉起盟主法:本實驗採用G 8 M 細胞株為實驗對象, 首先進行G 8 M 細胞株內室主氯基硫( gl utath ion e 'GS H ) 含量之測定, 接著以Lowry method 測定細胞內蛋白質之濃度,然後以放射線照射及/或8 S 0( D ' L-buthionine-S ' R-s ul fox i m i n e ) 處理細胞, 研究在有氯及缺氧環境下, 不同劑量放射線照射及/或8 S0處理後細胞G SH含量改變之情形。 益墨G 8 M 細胞內G S H 之濃度( [GS H]) 約為9 , 9 3 ± 0.46 (ugj mg of cellular p rote巾, 以下單位相同故省略) ,經過2 - 1 4 Gy 之放射線照射後不同時間, 隨著放射劑量的增加[GS H ] 有上升的趨勢。細胞經8S0處理後之濃度為8.7 8 ± 0.05 ' 經8S0 處理及放射線照射後不同時間之[ G S H ]隨著放射線劑量之增加也有上升的趨勢。缺氮G 8 M細胞內[ G S H ]約為3 2 . 4 5 ± 0.25 ' 經8 S 0 處理後之濃度為13.36 ±0.47 ' 經放射線照射/或8S0處理及放射線照射後不同時間之[ GS H ]隨著照射劑量之增加多有升高的現象。有氮之G 8M細胞經8S0處理後細胞內[ GS H ] 有下降之情形, 但是缺氧G 8M細胞只有在照射後立即分析這一組[ G S H ]減少,經過24及48小時再分析之兩組[ G S H ] 無明顯變化。除了在有氧狀態照射後立即分析組,其餘組之細胞經放射線照射後,隨著放射劑量的增加[ G S H ] 有上升的趨勢,且此上升趨勢經過8 S 0 處理後更為明顯。此外,放射線照射後之G 8 M細胞內( G S H )變化與有氧或缺氧狀態無顯著關係。 益益: I一l有氧G 8 M細胞經8S0處理後細胞內[GSH]有下降之情形,但是缺氧G8M細胞只有在照射後立即分析這一組[GSH]減少。(二)放射線照射後之G8M細胞內(GSH)變化與有氧或缺氧狀態無顯著關係。但)一般而言,G8M細胞經放射線照射後,隨著放射劑量的增加[GSH]有逐漸上升的趨勢,且此上升趨勢經8S0處理或在照射後24-48小時更為明顯。此種[GSH]逐漸上升可能表示隨著放射劑量增加細胞修復愈不明顯且輻射生物效應亦愈加顯著。
Purpose: To eva luate the efficacy of radi ation and BSO on the GSH concentration of GBM cells. Materials and Methods: Exponentially growing cultures of GBM/840 I cell s were used for this study. The GSH concentrations in GBM cells treated with radi ation and/or BSO (D. L-buthionin e-S. R-sulfo ximine) were determined. Cells were irradi ated with dosage ranged from 0 to 14 Gy with gamma radiation using a cobalt 60 machine. The concentra tion of GSH in irradiated GBM cells treated with or without BSO was evaluated under aerobic and hypoxic conditions. Results: The intracellul ar concentration of glutathione ([GSH]) was 9.93 ± 0.46 ug/mg of cellular protein. In general. the [GSHj increased gradually after irradi ated by cobalt-60 with 2 - 14 Gy. The [GSHI in GBM cells were 8.78 ± 0.05 after treated with BSO. Also, [GSHj increased gradually after irradi ated from low to high dose of irradiation. The [GSHI in hypoxi c GBM cell s were 32.45 ± 0.25. It was 13.36 ± 0.47 after treated with BSO. In general, the cellul ar [GSHj afte r irradiated from low to high dose of radi ation increa sed gradually no matter treated with BSO or not. BSO is able to deplete the intracellular [GSHj but this is not significa nt in hypo xic cells 24 and 48 hours after treatment. Change of intracellular [GSH j is also not closely related with the present of oxygen or not. Concl usion: ( I) . In aerobic gro up, the GSH concentrations in GBM cells decrease after treated with BSO. In hypoxic group, the GSH concentration decreased only in cells analyzed immed iately after irradiation. (2). There is no significant association between GSH co ncentrations of irradi ated GBM cells and various oxygenated conditions. (3). In general, GSH co ncentration increa se in proportional to radia tion dose . Thi s is more ev ident after treated the ce lls with BSO or 24-48 hours after irradi ation . Gradually increased concentration of GSH repre sent s less repair of the damaged cells from radiation and more biologic effec ts in accordance with high radiati on dose.
